A Comparative study on Determination of Lymphocyte Differential Count use the Manual Method Coulter, Counter-S plus Ⅱ and Diff₃50 System

Abstract
|
|
|
A comparision of analytical method for the determination of lymphocyte differential count was made between manual method, Coulter Counter S plus ll and Diff3 50 system.
The author evaluated reproducebility as well as correlation between three methods.
The results are as follow
1. The mean value of lyinphocrte percentage of peripheral blood from one healthy person were 23.3% by manual method,24.9% by coulter counter and 24.9% by Diff3 50 srstem.
2. Inter-assay variation (N : 20) represented by coefficient of variation was 22.85 by manual method,2.8% by coulter counter and 21.3% by Diff3 50 system.
3. A correlaton tot·ween three method(N : 135) was imade.
a. A correlation between manual and coulter counter.
R= 0.9531 and linear regression Y= 0.911x+2.7594
b. A correlation between coulter counter and Diff3 50 system.
R=0.9471 linea regression Y=0.942x+0.628
c. A correlation between manual and Diff3 50 system.
R=0.9026 linen regression Y=0.866x+2.821
4. Coulter counter-S plus ll system has a good correlation with manual or Diff3 50 system and very useful screening test for determination of lymphocyt percentage.
